目的:探讨在中小强度运动应激下右心室肌蛋白质组的差异表达及其规律,初步筛选与运动健康相关的备选目标蛋白质。方法:将20只SD雄性大鼠随机分为对照组和实验组,以跑台运动的方式,复制递增运动负荷至24 m/min的中小强度运动实验动物模型;提取右室肌组织的全蛋白进行双向凝胶电泳分离,运用ImageMaster 2D Platinum图像分析软件对2-DE胶进行分析。结果:2-DE图谱上,对照组检测蛋白质点(437±17)个,实验组检测(448±19)个;运动后47个蛋白质点发生了较明显的变化,其中,1个蛋白质点消失,表达量上调2倍以上的点20个,下调至50%以下的15个,上调在5倍以上的点8个,下调至20%以下的3个。结论:4周中小强度运动后,大鼠右心室肌蛋白质组发生了显著的变化,初步筛选出12个备选目标蛋白质点,为研究运动健康与运动康复具有重要意义的新蛋白奠定前期工作基础。
OBJECTIVE: To explore the differential expression and regularity of proteomics of right ventricular muscle in small and medium-intensity exercise stress, and to screen preliminary alternative target proteins related to exercise and health. Methods: Twenty male Sprague-Dawley rats were randomly divided into control group and experimental group. The treadmill exercise was used to replicate the small and medium-intensity exercise animal model with increasing exercise load to 24 m / min. The two-dimensional gel electrophoresis separation, the use of ImageMaster 2D Platinum image analysis software 2-DE glue analysis. Results: On the 2-DE map, the number of protein spots in the control group (437 ± 17) and the experimental group (448 ± 19) showed significant changes after the exercise, in which one protein spot disappeared , 20 up-regulated by more than 2 folds, 15 down-regulated to below 50%, 8 up-regulated by more than 5 folds and down to 3 below 20%. CONCLUSION: After 4 weeks of moderate and small intensity exercise, the proteomics of right ventricular muscle of rats has been significantly changed. Twelve candidate target proteins were initially screened, which laid the foundation for the preliminary study on new proteins of great significance for sports health and sports rehabilitation .
