目的探讨海昆肾喜对阿霉素肾病大鼠肾组织结缔组织生长因子(connectivetissuegrowthfactor,CT-GF)蛋白及其mRNA的影响。方法将54只大鼠随机分为6组,正常组、假手术组、模型组、苯那普利组和海昆肾喜小剂量组与大剂量组。采用单侧肾切除阿霉素肾病模型。术后10周处死大鼠,观察肾组织病理改变,并应用免疫组织化学方法和原位杂交法检测肾组织CTGF及其mRNA的表达。结果正常组及假手术组肾小管上皮细胞、肾间质细胞胞浆内有少量CTGF及其mRNA阳性反应物;模型组肾小管上皮细胞、肾间质细胞内的CTGF及其mRNA在细胞浆内呈强阳性表达;海昆肾喜小剂量组、大剂量组表达较模型组显著减少(P<0.05)。结论海昆肾喜可抑制CTGF蛋白及其mRNA的过度表达,进而减缓肾纤维化的病程进展。
Objective To investigate the effect of Haikun Shenxi on renal connective tissue growth factor (CT-GF) protein and its mRNA in rats with adriamycin-induced nephropathy. Methods 54 rats were randomly divided into 6 groups: normal group, sham operation group, model group, benazepril group and Haikun Shenxi low dose group and large dose group. Unilateral nephrectomy with adriamycin nephropathy model was used. The rats were sacrificed 10 weeks after operation and the pathological changes of the kidneys were observed. The expression of CTGF and its mRNA was detected by immunohistochemistry and in situ hybridization. Results There was a small amount of CTGF and its mRNA positive reaction in the cytoplasm of tubular epithelial cells and renal interstitial cells in normal group and sham operation group; CTGF and mRNA in renal tubular epithelial cells and renal interstitial cells of model group were in cytoplasm. Strong positive expression; Haikun Shenxi small dose group, high dose group expression was significantly reduced compared with the model group (P <0.05). Conclusion Haikun Shenxi can inhibit the overexpression of CTGF protein and its mRNA, and thus slow down the progression of renal fibrosis.
