为评价胸腺基质细胞区的辐射效应,作者以平均能量1MeV裂变中子或300kVP X线分割全身照射新出生的CBA/H小鼠,剂量分别为6.88和15.00Gy。照后20min内取出胸腺,分二叶分别移植于C3H裸鼠肾被膜下。移植后1~2个月,再取出该胸腺移植物,计量其体积,然后以免疫细胞化学法观察移植物内T细胞和基质细胞的分布及结构,并以流动细胞荧光计分析实验动物脾细胞的T细胞亚群。
To assess the radiation effects of the thymus stromal cell area, newborn CBA / H mice were irradiated whole-body with a mean energy of 1 MeV fission neutrons or 300 kVP X-rays at doses of 6.88 and 15.00 Gy, respectively. According to 20min after removal of the thymus, sub-two leaves were transplanted in the kidney capsule of C3H nude mice. After 1 to 2 months after transplantation, the thymus graft was removed and its volume was measured. Then the distribution and the structure of T cells and stromal cells in the graft were observed by immunocytochemical method. The splenocytes of experimental animals were analyzed by flow cytometer T cell subsets.
