目的观察天花粉蛋白经由TCR/CD3介导的信号传递途径抑制T细胞活化增殖的效应。方法用各种不同的剌激信号来激活外周血淋巴细胞,然后观察天花粉蛋白对其增殖抑制的影响。接着测定天花粉蛋白对CD3单抗剌激后T淋巴细胞内一些信号传导物质变化的影响。结果天花粉蛋白抑制经由TCR/CD3信号传导途径诱发的淋巴细胞增殖。进而发现细胞内游离钙离子的升高,蛋白激酶C的转位和细胞内蛋白酪氨酸磷酸化受天花粉蛋白的抑制。结论天花粉蛋白可通过TCR/CD3的信号传导途径抑制T细胞增殖。
Objective To observe the effect of trichosanthin inhibiting the activation and proliferation of T cells via TCR/CD3-mediated signaling pathway. Methods Activate peripheral blood lymphocytes with various stimulation signals, and then observe the effect of trichosanthin on its proliferation inhibition. Next, the effect of trichosanthin on the changes of some signaling substances in T lymphocytes stimulated by CD3 monoclonal antibody was determined. As a result, trichosanthin inhibits lymphocyte proliferation induced via the TCR/CD3 signaling pathway. Furthermore, it was found that the increase of intracellular free calcium, the translocation of protein kinase C, and the intracellular protein tyrosine phosphorylation were inhibited by the trichosanthin protein. Conclusion Trichosanthin inhibits T cell proliferation through TCR/CD3 signaling pathway.
