一、测定的原理 TTC又叫红四氮唑,全名2,3,5,—氯代三苯基四氮唑,学名Tipheny Tetrazolium Chloride,(北京化工厂生产)为白色至黄白色粉末,是一种氧化还原剂,易溶于水,更易溶于醇等有机溶剂。其染色原理是:TTC在有生活力种子胚组织活细胞中脱氢酶的作用下,被还原成红色稳定的2,3,5——三苯基甲脂(TTCH),而无生活力的种子就没有这种反应,从而区别出种子有无生活力。 二、测定的方法 1、配制溶液:用蒸馏水(或pH6~7的洁净水)把TTC配成0.5~1%浓度的溶液。
