目的:了解急性早幼粒细胞白血病(APL)的早幼粒细胞白血病锌指基因(PLZF)和17号染色体上的RARα基因与APL发生的关系。方法:应用PCR克隆方法,缺失PLZF/RARα的PLZF不同区域,构建了8个PLZF/RARα的突变体进行细胞转染。结果和结论:证实了PLZF/RARα对野生型RARα有明显的显性负作用,而POZ区域(即氨基酸1~120)对PLZF/RARα的显性负作用的形成是至关重要的,2个锌指结构可能通过与POZ区域的空间构型的相互作用而在显性负作用中起一定作用。
OBJECTIVE: To investigate the relationship between the promyelocytic leukemia (APL) pronuclear leukemia zinc finger gene (PLZF) and RARα gene on chromosome 17 and the occurrence of APL. METHODS: Eight PLZF/RARα mutants were constructed by PCR cloning and PLZF/RARα deletion in different regions. Results and Conclusion: It was confirmed that PLZF/RARα has a significant negative effect on wild-type RARα, while the POZ region (ie, amino acids 1 to 120) is critical to the formation of a dominant negative effect of PLZF/RARα, 2 The zinc finger structure may play a role in the dominant negative effect through the interaction with the steric configuration of the POZ region.
