This article describes the effect of human serum with or without Epstein-Barr virus (EBV) antibodies on the virus proliferation in P3HR-1 cells. Cell culture with EBV-containing sera resulted in a decrease in the amount of infectious and virion DNA in the supernatant and a decrease in intracellular EBV DNA. The reduction of EBV DNA for this cellular dependence depends on the concentration of serum added and the incubation time. EBV DNA was detected by gel electrophoresis and DNA hybridization and found that extrachromosomal EBV DNA existed in two forms within the virus-replicating cells; additional CCC-DNA (covalently closed circular DNA and virion-associated L-DNA (Linear-DNA).
