目的探讨邻苯二甲酸单丁酯(mono-n-butyl phthalate,MBP)对小鼠睾丸氧化损伤的影响。方法 36只性成熟的雄性Balb/c小鼠随机分为4个MBP染毒组(25、50、100、200 mg/kg)和1个空白对照组、1个溶剂对照组(食用油),每组6只,灌胃14 d后观察睾丸的组织形态学损伤;检测睾丸组织的活性氧(ROS)、丙二醛(MDA)、还原型谷胱甘肽(GSH)的含量。结果 100和200 mg/kg组小鼠睾丸组织不同发育时期生殖细胞的数量减少;随着MBP染毒剂量的升高,睾丸组织ROS含量与MDA含量逐渐上升,GSH含量逐渐降低;100和200 mg/kg组上述指标与对照组比较,差异有统计学意义(P<0.05)。结论较高剂量的MBP会引起小鼠睾丸细胞的氧化损伤。
Objective To investigate the effect of mono-n-butyl phthalate (MBP) on the oxidative damage of testis in mice. Methods Thirty-six male Balb / c mice were randomly divided into 4 MBP groups (25, 50, 100, and 200 mg / kg), 1 blank control group, 1 solvent control group Six rats in each group were sacrificed for 14 days. Morphological changes of testis were observed. The contents of reactive oxygen species (ROS), malondialdehyde (MDA) and reduced glutathione (GSH) in testis tissue were measured. Results The numbers of germ cells in testis tissue of 100 and 200 mg / kg groups decreased at different developmental stages. The ROS and MDA contents in testis tissue gradually increased and the content of GSH decreased gradually with the increase of MBP dose. The levels of 100 and 200 mg / kg group compared with the control group indicators, the difference was statistically significant (P <0.05). Conclusion Higher doses of MBP cause oxidative damage in testicular cells of mice.
