目的探讨镧(lanthanum,La)对大鼠大脑皮质星形胶质细胞(astrocyte,AS)的损害效应及对GRP-78、IRE1、JNK、Caspase-3和c PARP表达的影响。方法 0、0.25、0.5和1.0 mmol/L La Cl3处理大鼠大脑皮质AS细胞24 h,然后用乳酸脱氢酶(lactic dehydrogenase,LDH)检测试剂盒测AS细胞LDH释放量,流式细胞仪测AS细胞凋亡情况,Western blot法测AS细胞GRP-78、IRE1、磷酸化的IRE1(phosphorylated IRE1,p IRE1)、JNK、磷酸化的JNK(phosphorylated JNK,p JNK)、Caspase-3和c PARP表达。结果 0.25、0.5和1.0 mmol/L La Cl3处理的大鼠AS细胞LDH释放量和细胞凋亡率显著高于对照AS细胞,且有剂量-反应关系。0.25、0.5和1.0 mmol/L La Cl3处理的大鼠AS细胞GRP-78、p IRE1、p JNK、Caspase-3和c PARP表达显著高于对照AS细胞,且随La Cl3处理AS细胞的剂量增加而升高,其中1.0 mmol/L La Cl3处理的大鼠AS细胞GRP-78、p IRE1、p JNK、Caspase-3和c PARP表达分别为对照AS细胞的2.60倍、2.96倍、3.93倍、3.38倍和2.93倍。结论 La对大鼠大脑皮质AS细胞的损害效应可能和GRP-78水平升高、IRE1和JNK磷酸化增加及Caspase-3、c PARP表达增强而致凋亡增加有关。
Objective To investigate the effect of lanthanum (La) on the damage of cerebral cortex astrocyte (AS) and the expression of GRP-78, IRE1, JNK, Caspase-3 and c PARP in rats. METHODS: Rat cerebral cortex AS cells were treated with 0, 0.25, 0.5 and 1.0 mmol / L La Cl3 for 24 h, then LDH release was measured by lactic dehydrogenase (LDH) kit. Flow cytometry AS cells were detected by Western blot. The expressions of GRP-78, IRE1, phosphorylated IRE1 (p IRE1), JNK, phosphorylated JNK (p JNK), Caspase-3 and c PARP expression. Results The LDH release and apoptosis rate of AS cells treated with 0.25, 0.5 and 1.0 mmol / L LaCl3 were significantly higher than that of control AS cells, and there was a dose-response relationship. The expressions of GRP-78, p IRE1, p JNK, Caspase-3 and c PARP were significantly higher in AS cells treated with 0.25, 0.5 and 1.0 mmol / L LaCl 3 compared with control AS cells, and the dose of AS cells treated with LaCl 3 was increased The expression of GRP-78, p IRE1, p JNK, Caspase-3 and c PARP in AS cells treated with 1.0 mmol / L LaCl3 were 2.60, 2.96, 3.93 and 3.38 Times and 2.93 times. CONCLUSION: The effect of La on the damage of AS cells in rat cerebral cortex may be related to the increase of GRP-78, the increase of IRE1 and JNK phosphorylation and the increase of the expression of Caspase-3 and c PARP.
