目的 预测广州地区人巨细胞病毒(HCMV)临床低传代UL144基因B细胞表位.方法 应用生物信息学方法,改进型自我优化预测结构(SOPMA),Garnier-Osguthorpe-Robson (GOR),人工神经网络预测法(HNN)预测二级结构,隐马尔可夫模型(TMHMM)预测跨膜结构,分别用Hopp&Woods、Zimmerman、Janin、Welling、Flexibility预测亲水性、极性、可及性、抗原性、柔韧性.结果 预测HCMV UL144编码产物包含176个氨基酸残基,等电点为8.97.二级结构预测结果显示:3-转角及无规则卷曲主要位于31-42、57-63、71-78、87-93、96-100、127-131氨基酸区域.TMHMM预测UL144氨基酸序列为一次跨膜蛋白,其跨膜区域为134-156氨基酸区域,胞外及胞内区域分别为:1-133、157-176氨基酸区域.亲水性位于39-47、108-117、124-131、157-164区域;表面可及性区域为22-30、39-50、94-103、105-132、156-164;极性区域位于17-37、39-54、69-77、81-89、94-104、106-136、156-164;抗原性区域位于21-26、97-102、107-125、127-136;柔韧性区域位于27-47、56-71、85-95、101-106、108-118、123-131、156-172.结论 综合各项参数,UL144基因106-125、94-102、21-30氨基酸区域抗原指数(AI)较高,预测该区域内或其附近为B细胞表位的位点.“,”Objective To predict human Cytomegalovirus (HCMV) clinical low-passage UL144 gene B cell epitope in Guangzhou area.Methods Bioinformatics method,improved self-optimization prediction method from alignment (SOPMA),Garnier-Osguthorpe-Robson (GOR),artificial neural network prediction (HNN) secondary prediction structure,TMHMM predictive transmembrane structure were used to predict hydrophilicity,polarity,accessibility,flexibility,and antigenicity by Hopp&Woods,Zimmerman,Janin,Welling,and Flexibility.Results The predicted HCMV UL144 encoded products contained 176 amino acid residues,and the isoelectric point was 8.97.The results of secondary prediction structure showed that β-turn and random coil were mainly located at 31-42,57-63,71-78,87-93,96-100,and 127-131 amino acid regions.TMHMM predicted that UL144 amino acid sequence was a transmembrane protein,134-156 amino acids formed a transmembrane region,the extracellular and intracellular regions were 1-133 amino acid region and 157-176 amino acid region,respectively.The hydrophilic property was located at 39-47,108-117,124-131,and 157-164 amino acid regions;the surface accessibility was located at 22-30,39-50,94-103,105-132,and 156-164 amino acid regions,the polarity was located at 17-37,39-54,69-77,81-89,94-104,106-136,and 156-164 amino acid regions,the antigenicity was located at 21-26,97-102,107-125,and 127-136 amino acid regions,the flexibility was located at 27-47,56-71,85-95,101-106,108-118,123-131,and 156-172 amino acid regions.Conclusion Among the parameters,the levels of antigen index (AI) of UL144 gene 106-125,94-102,and 21-30 amino acid regions are high,and the sites of B cell epitopes in or near the region are predicted.
