目的 探讨急性髓系白血病 (AML)患者端粒酶活性、端粒长度的变化及意义。方法 采用TRAP ELISA PAGE法测定端粒酶表达活性 ,Southernblot方法测定端粒长度。结果 AML患者端粒酶活性 [吸光度 (A)值为 2 .2 98± 1.0 5 9]较正常对照 (A值为 0 .3 87± 0 .5 98)显著增高 ,同时其平均端粒长度 [( 7.6± 2 .1)kb]较正常对照 [( 9.3± 1.9)kb]明显缩短。端粒长度的变化主要发生在端粒酶活性高表达的白血病细胞中。结论 AML细胞端粒酶活化可能与其端粒缩短有关。端粒的丢失可导致染色体的失稳 ,激活端粒酶依赖的端粒机制 ,在白血病的发生中起着重要作用。
Objective To investigate the changes of telomerase activity and telomere length in patients with acute myeloid leukemia (AML) and its significance. Methods Telomerase activity was determined by TRAP ELISA PAGE and telomere length was determined by Southern blotting. Results The telomerase activity of AML patients (absorbance (A) value was 2.298 ± 1.059) was significantly higher than that of the normal control (A value was 0.087 ± 0.598), and mean telomere length [ (7.6 ± 2.1) kb] was significantly shorter than the normal control (9.3 ± 1.9) kb. Changes in telomere length mainly occur in leukemic cells with high telomerase activity. Conclusion The telomerase activation in AML cells may be related to its telomere shortening. Loss of telomeres can lead to chromosomal instability, activation of telomerase-dependent telomere machinery, plays an important role in the development of leukemia.
