目的:探求LAK细胞对白血病疗效的监测手段。方法:用流式细胞仪测定5例成人脾淋巴因子激活的杀伤细胞(LAK细胞)和21例正常人外周血淋巴细胞的DNA指数(DI)及增殖周期。结果:人脾LAK细胞的增殖活力明显高于未激活的脾淋巴细胞和外周血淋巴细胞。培养(1~14d)期间人脾LAK细胞的增殖活力高,并且稳定。在人脾LAK细胞中,未找出非整倍体细胞,DI指数在正常范围。结论:流式细胞仪可用于判断人脾LAK细胞对白血病的疗效。
Objective: To explore the means of monitoring leukemia efficacy in LAK cells. Methods: The DNA index (DI) and proliferation cycle of 5 spleen lymphokine activated killer cells (LAK cells) and 21 normal human peripheral blood lymphocytes were determined by flow cytometry. Results: The proliferation activity of human spleen LAK cells was significantly higher than that of unactivated spleen lymphocytes and peripheral blood lymphocytes. The proliferation activity of human splenic LAK cells was high and stable during the period of 1 to 14 days. In human spleen LAK cells, aneuploid cells were not found and the DI index was in the normal range. Conclusion: Flow cytometry can be used to determine the efficacy of human spleen LAK cells on leukemia.
