目的:优选忍冬藤叶中绿原酸的分离纯化工艺条件。方法:以绿原酸含量为指标,通过单因素试验考察聚酰胺树脂的静态和动态吸附、解吸性能,并考察上样量、水洗用量、洗脱剂种类和用量;以绿原酸纯度和保留率为指标,采用L9(34)正交试验考察洗脱流速、洗脱剂浓度和用量对忍冬藤叶中绿原酸的分离纯化工艺的影响。结果:优选的分离纯化工艺为绿原酸上样量10 mg·g-1树脂,用2 BV水洗除杂,加3 BV 65%乙醇洗脱,洗脱流速0.4 BV·h-1,收集洗脱液。结论:优选的工艺合理、稳定可行,可为工业化分离纯化绿原酸提供参考。
Objective: To optimize the conditions for the isolation and purification of chlorogenic acid in honeysuckle leaves. Methods: The content of chlorogenic acid was used as an index to investigate the static and dynamic adsorption and desorption properties of polyamide resin by single factor experiment. The amount of sample, the amount of washing, the type and amount of eluent were investigated. Rate as an indicator, using L9 (34) orthogonal test elution flow rate, eluent concentration and amount of chlorogenic acid in honeysuckle leaves of the separation and purification process. Results: The optimum conditions for the isolation and purification of chlorogenic acid were as follows: 10 mg · g-1 chlorogenic acid resin, 2 BV water wash, 3 BV 65% ethanol elution, elution flow rate 0.4 BV · h-1, De-liquid. Conclusion: The optimized process is reasonable, stable and feasible, which can provide a reference for the industrialization of isolation and purification of chlorogenic acid.
