Argonaute2(Ago2)蛋白在生物体内广泛表达,是RNA诱导沉默复合体的核心成分,具有核糖核酸内切酶活性,可通过促进微RNA的成熟并调节其生物合成及功能,进而抑制靶基因的表达,在多种病理生理过程中起关键性作用。 Ago2表达水平变化或功能失调时,可作为抑癌或致癌基因参与多种肿瘤的发生、发展,与肿瘤细胞的增殖与分化、新生血管的发生、对缺氧应激的耐受性等密切相关。随着研究的深入,Ago2有望成为肿瘤诊断与治疗的新靶点,该文就Ago2在肿瘤中的研究进展予以综述。“,”Argonaute 2(Ago2) protein with endoribonuclease activity is the core of RNA-induced silen-cing complex,it has been identified to be widely expressed in human somatic cells and involved in post-tran-scriptional gene silencing by regulating the biogenesis and function of small RNAs such as microRNA,and thus plays key roles in physiological and pathological processes.Ago2 expression level changes or dysfunction can make it act as a tumor suppressor or oncogene to influence carcinogenesis,such as tumor cell proliferation and differentiation,angiogenesis and stress response.Therefore,Ago2 has the potential value of becoming a new marker to the diagnose and therapy of human cancer.Here is to make a review of the research progress of Ago2 in tumors.
