为探讨紫草素在裸鼠体内抗滋养细胞肿瘤的机制,应用电化学发光法,计算瘤重抑制率,检测紫草素对绒癌的抑制作用;同时应用HE染色及免疫组化法检测用药组及溶媒对照组肿瘤组织的病理改变及抑制凋亡基因bcl-2蛋白的表达。结果:①紫草素组的平均瘤重明显低于溶媒对照组(P<0.05),对肿瘤的抑制率大于30%;②紫草素组从停药第1天开始人绒毛膜促性腺激素(β-HCG)急剧下降,至第3天最低降至2000IU/L左右,显著低于溶媒对照组,差异有非常显著意义(P<0.01);③紫草素能够使肿瘤组织细胞发生大面积坏死,坏死组织内可见细胞核凝固或碎裂,最后细胞核溶解消失;④紫草素组肿瘤组织细胞bcl-2蛋白表达明显减少。结论:紫草素能够有效地诱导绒癌裸鼠移植肿瘤细胞凋亡和坏死,明显抑制β-HCG的分泌;Bcl-2蛋白的表达下调是紫草素抗绒癌作用的重要机制。

CONCLUSION: Shikonin can effectively induce apoptosis and necrosis of tumor cells in nude mice bearing choriocarcinoma and significantly inhibit the secretion of β-HCG. The down-regulation of Bcl-2 protein expression is an important mechanism of anti-choriocarcinoma effect of shikonin.
