目的探讨三维适形调强放疗(intensity modulated radiationtherapy,IMRT)对子午沙鼠骨细粒棘球蚴原头节的杀灭作用及其对葡萄糖-6-磷酸酶表达的抑制作用。方法双股骨骨细粒棘球蚴病子午沙鼠40只,左侧股骨骨细粒棘球蚴病区给予总剂量为40Gy的IMRT放疗,右侧股骨骨细粒棘球蚴病区仅给予假照射作为阴性对照。于放疗结束1周后取鼠左、右侧股骨细粒棘球蚴原头节行伊红染色检测原头节坏死率,采用硝酸铅法检测葡萄糖-6-磷酸酶的活性变化。结果 IMRT作用1周后,可见子午沙鼠左侧股骨骨细粒棘球蚴原头节整体结构消失,出现深蓝着色;左侧股骨骨细粒棘球蚴原头节坏死率((71.26±21.85)%)高于右侧((5.93±5.77)%),差异有统计学意义(P<0.05);子午沙鼠左侧股骨骨细粒棘球蚴原头节的葡萄糖-6-磷酸酶活性较右侧明显减弱,葡萄糖-6-磷酸酶产物光密度值(0.271 2±0.104 2)明显低于右侧(0.642 9±0.398 5),差异有统计学意义(P<0.05)。结论 IMRT可促进鼠骨细粒棘球蚴原头节的死亡,其作用机制可能是通过改变葡萄糖-6-磷酸酶的活性而实现。
Objective To investigate the killing effect of three-dimensional intensity modulated radiation therapy (IMRT) on protoscoleces of Echinococcus granulosus and inhibition of glucose-6-phosphatase expression. Methods 40 females of double-stalk granulomatous echinococcus granulosus were given IMRT radiation with a total dose of 40 Gy to the left femur. The right femoral granulosomal echinococcosis area was only given fake Irradiation served as a negative control. One week after the end of radiotherapy, the protoscoleces necrosis rate was detected by eosin staining of the left and right femoral hydatid cysts. The activity of glucose-6-phosphatase was detected by the lead nitrate method. Results After IMRT for 1 week, the whole structure of the protoscolei of Echinococcus granulosus in the left femoral gerbils disappeared and the dark blue staining appeared. The necrosis rate of the protoscoleces of the hydatid cyst of the left femur ((71.26 ± 21.85 ) Were higher than the right (5.93 ± 5.77)%, the difference was statistically significant (P <0.05). The glucose-6-phosphatase activity of protoscoleces in the left femur bone of Meriones unguiculatus (P <0.05). The optical density of glucose-6-phosphatase (0.271 2 ± 0.104 2) was significantly lower than that of the right (0.642 9 ± 0.398 5), the difference was statistically significant (P <0.05). Conclusion IMRT can promote the death of protoscoleces of Echinococcus granulosus, and its mechanism may be through changing the activity of glucose-6-phosphatase.
