目的以长白山林蛙卵为原料,利用超声波辅助提取雌二醇。方法在单因素试验的基础上,采用Design-Expert软件中的Central-Composite中心组合设计,确定林蛙卵提取雌二醇的最佳工艺条件,建立回归数学模型。结果曲面回归方程拟合性好,雌二醇提取的最佳工艺条件:提取时间60min;乙醇浓度为80%;超声功率250W;提取温度70℃;料液比1∶10。在此试验条件下,采用酶联免疫分析(ELISA)检测得到雌二醇含量为62.39 ng/L。结论实验为林蛙卵资源的综合利用和天然雌激素的开发与应用提供了理论依据。
Objective To use Changbai Mountain Rana egg as raw material and use ultrasonic to extract estradiol. Methods Based on the single factor experiment, the Central-Composite center design in Design-Expert software was used to determine the optimum conditions for the extraction of estradiol from Rana chensinensis eggs, and a regression mathematical model was established. Results The regression equation of the surface fitting well, the best conditions for the extraction of estradiol: extraction time 60min; ethanol concentration 80%; ultrasonic power 250W; extraction temperature 70 ℃; Under this test condition, the estradiol content was 62.39 ng / L by enzyme-linked immunosorbent assay (ELISA). Conclusion The experiment provided the theoretical basis for the comprehensive utilization of Rana eggs and the development and application of natural estrogen.
