目的研究生殖器疱疹患者外周血中HSV Ⅱ抗原特异性CD8+细胞毒性T细胞(CTL)在发病中的作用。方法采用荧光标记MHCⅠ类分子-多肽五聚体技术,以流式细胞仪定量检测20例生殖器疱疹患者初发和复发双份外周血中HSV Ⅱ抗原特异性CD8+细胞毒性T细胞。并进行比较分析。结果生殖器疱疹患者初发和复发双份外周血中HSV Ⅱ抗原特异性CD8+细胞毒性T细胞百分数分别为0.73%±0.29%和0.35%±0.24%,正常对照组为0.31%±0.27%,初发组与复发组、正常对照两组结果相比较P<0.001;复发组和正常对照组结果相比较P>0.05。结论初发生殖器疱疹患者外周血HSV Ⅱ抗原特异性CD8+细胞毒性T细胞活性增强,复发时患者CTL的活性减弱。五聚体技术具有快速、敏感、高效和特异的特点。
Objective To study the role of HSV Ⅱ antigen-specific CD8 + cytotoxic T lymphocytes (CTL) in the pathogenesis of genital herpes in patients. Methods HSV Ⅱ antigen-specific CD8 + cytotoxic T cells in 20 peripheral blood samples from 20 patients with genital herpes were detected by flow cytometry using fluorescence-labeled MHC class Ⅰ-peptide pentamer technique. And comparative analysis. Results The percentages of HSV Ⅱ antigen-specific CD8 + cytotoxic T cells in both genital herpes inpatients and relapsed patients were 0.73% ± 0.29% and 0.35% ± 0.24%, respectively, and those in the normal control group were 0.31% ± 0.27% P <0.001 in the group and recurrence group and normal control group, P> 0.05 in the group of recurrence group and the normal control group. Conclusions The activity of HSV Ⅱ antigen-specific CD8 + cytotoxic T cells in peripheral blood of patients with genital herpes is increased, and the CTL activity of patients with recurrent genital herpes is weakened. Pentamer technology is fast, sensitive, efficient and specific.
