应用反转录-聚合酶链式反应(RT-PCR)及限制性片段长度多态性(RFLP)和单链构象多态性(SSCP)分析技术,快速检测我国水稻条纹病毒(Rice stripe virus, RSV) RNA4基因间隔区(intersenic region,IR)的变异。 IR经 RT-PCR扩增后得到一段长约 680 bp的片段, RSV 7个分离物之间片段大小相同,应用AluI、 NdeI在它们之间检测到 RFLP,其中 BS、 JD、 LY、 YL分离物完全一致,而 SQ、 PJ、 JN分离物各不相同。 RSV 7个分离物的 SSCP泳动带型证实了 RFLP分析结果。这是我国RSV分子变异的首次报道。
The rapid detection of rice stripe virus (Rice stripe virus) in China by reverse transcription-polymerase chain reaction (RT-PCR) and restriction fragment length polymorphism (RFLP) and single strand conformation polymorphism (SSCP) RSV) RNA4 gene interspace (intersenic region, IR) variation. IR amplified a fragment of about 680 bp by RT-PCR. The fragment sizes of RSV 7 isolates were the same. RFLP was detected among them using AluI and NdeI, among which BS, JD, LY and YL isolates Exactly the same, while SQ, PJ, JN isolates vary. The SSCP bands of the RSV 7 isolates validated the RFLP analysis. This is the first report on the molecular variation of RSV in China.
