目的:探讨降压宝蓝片含药血清对过氧化氢(H2O2)诱导人脐静脉血管内皮细胞(HUVECs)损伤的保护作用。方法:体外培养HUVECs,分为空白组、模型组、降压宝蓝片含药血清低、中、高浓度组(含量血清体积分数为5%,10%,15%),显微镜下观察细胞形态,用噻唑蓝(MTT)法检测HUVECs活力,微板法检测上清液中乳酸脱氢酶(LDH)的活性及一氧化氮(NO)的含量,ELISA法检测内皮素-1(ET-1),组织型纤溶酶原激活物(t PA),组织型纤溶酶原激活物抑制剂(PAI-1)的含量。结果:与空白组比较,模型组细胞活力显著降低,上清液中LDH活性明显增高,ET-1和PAI-1含量显著增加,NO和t PA含量显著下降(P<0.01);与模型组比较,降压宝蓝片中、高浓度组细胞活力显著升高,LDH活性显著减低,ET-1和PAI-1含量显著减少,NO和t PA显著升高(P<0.05,P<0.01)。结论:降压宝蓝片含药血清对H2O2诱导的HUVECs损伤具有显著地保护作用。
Objective: To investigate the protective effect of Jianbo Bao Lan Tablet containing serum on the injury of human umbilical vein endothelial cells (HUVECs) induced by hydrogen peroxide (H2O2). Methods: HUVECs were cultured in vitro and divided into blank group, model group, Jianbo Bao Lan Tablet containing low, medium and high serum concentration (content of serum volume fraction of 5%, 10%, 15%) under a microscope to observe the cell morphology, The activity of lactate dehydrogenase (LDH) and nitric oxide (NO) in supernatant were detected by MTT assay and the activity of endothelin-1 (ET-1) , Tissue-type plasminogen activator (t PA), tissue-type plasminogen activator inhibitor (PAI-1) content. Results: Compared with the blank group, the viability of model group decreased significantly, the activity of LDH in the supernatant increased significantly, the contents of ET-1 and PAI-1 increased significantly and the content of NO and t PA decreased (P <0.01) Compared with the control group, the cell viability in high concentration group was significantly increased, the LDH activity was significantly decreased, the content of ET-1 and PAI-1 was significantly decreased, NO and t PA were significantly increased (P <0.05, P <0.01). Conclusion: The antihypertensive Baolan tablets containing serum have a significant protective effect on H2O2-induced HUVECs injury.
