Result It was observed that MEK 5 specific inhibitor BIX02188inhibited the formation of neurospheres and specifically blocked ERK 5phosphorylation but not ERK 1/2 phosphorylation, Nerve growth factor(NGF) significantly increased the phosphorylation of ERK 5 and acted asactivator following inhibition by BIX02188. NGF activated phosphorylationofboth ERK 1/2 and ERK. After be treated with 20 μM PD98059 and 15 μMBIX20188, and then activated by fluoxetine, there was significance increasephosphorylation ofboth ERK 1/2 and ERK 5.
Conclusion Fluoxetine activates ERK 5 is essential for the proliferationofNSCs derived from mouse embryonic hippocampus. Taking these resultstogether, fluoxetine not only played important role along MEK/ERK 1/2 butalso MEK/ERK 5 pathway aside from MEK/ERK 1/2 interference.
