大白菜(Brassica compestris L.ssp.pekinensis)是我国重要的蔬菜作物。近年来,育种材料单一已成为育种取得突破性进展的瓶颈,为了选育稳定的二体异附加系,以大白菜—结球甘蓝单体及双单体异附加系为试材,采用常规压片、核型分析以及SSR标记技术从其自交后代中鉴定出4个二体异附加系,并对二体异附加系的形态学、细胞学及生殖特性等方面进行了研究。为大白菜—结球甘蓝异代换系、易位系的创建提供了实践和理论基础,并为研究基因的互作、定位和表达以及加速大白菜遗传改良奠定了基础。主要研究结果如下:1.利用根尖有丝分裂中期计数法,从大白菜—结球甘蓝1号和6号双单体异附加系、3号、7号、9号单体异附加系中筛选出2n=22的植株。其所占比例分别为7.89%、6.06%、4.65%、5.26%。并对所得植株进行染色体核型分析,初步确定为附加结球甘蓝1号、3号、7号和9号染色体的大白菜—结球甘蓝二体异附加系,分别命名为AC1d、AC3d、AC7d和AC9d。2.对AC1d、AC3d和AC7d三个大白菜—结球甘蓝二体异附加系植株花粉母细胞减数分裂进行观察。结果表明,终变期以11Ⅱ联会形式为主,所占比率分别为90.10%、60.8%、71.1%;中期I大部分排列于赤道板,少数以两条染色体游离于赤道板外的形式存在;后期Ⅰ分离方式多样,但基本以11-11形式分离;后期Ⅱ以11-11-11-11为主,分别占所有分离方式的83.50%、77.6%、69.9%。3.通过对AC1d、AC3d和AC7d 3个二体异附加系的植株花粉总量、花粉整齐度、花粉生活力进行统计,并与二倍体大白菜进行对比。结果表明,异附加系的花粉量、花粉活力与二倍体白菜相比明显减少,达到了显著差异,而不同的二体异附加系之间不存在显著差异;二倍体大白菜的花粉整齐度与AC3d之间的差异性不显著,而与其他的异附加系相比,明显要高,差异性显著。4.表型鉴定表明,AC1d营养生长时期,长势、外形等田间性状与二倍体大白菜没有明显的差异;AC1d、AC3d、AC7d生殖生长时期具有部分结球甘蓝的特征,如抽薹晚,花瓣较大,花色略白等特征;3个异附加系之间形态特征各异,结籽率极低。5.分别选取结球甘蓝03、02、08、06连锁群特异SSR引物,对异源三倍体杂交种、亲本大白菜、结球甘蓝、单体及二体异附加系进行分子鉴定。结果表明,各对引物在大白菜—结球甘蓝单体及二体附加系扩增结果与结球甘蓝及异源三倍体杂交种一致,而二倍体大白菜上没有相应的扩增产物。说明所筛选的AC1d、AC3d、AC7d和AC9d大白菜—结球甘蓝二体异附加系外源染色体分别与结球甘蓝的03、02、08、06连锁群相对应。
