目的:研究南方红豆杉枝叶及其愈伤组织萃取紫杉醇后剩余部分对α-糖苷酶活力的影响。方法:将萃取紫杉醇后剩余部分分别加入α-淀粉酶、α-麦芽糖酶、α-葡萄糖苷酶不同微量反应体系中,分别用Bernfeld法、PNPG法、GOD法测定其酶活性,并采用Dixon作图法研究其动力学性质。结果:枝叶及其愈伤组织对α-麦芽糖酶和α-葡萄糖苷酶有明显的抑制作用,与拜唐苹的作用一致,其抑制存在量效关系,抑制α-麦芽糖酶活性[IC50=(218.49±18.24)mg/mL和(91.04±5.43)mg/mL]呈可逆性、竞争性抑制,抑制α-葡萄糖苷酶活性[IC50=(7.99±1.57)mg/mL和(19.87±2.52)mg/mL]呈不可逆性、竞争性抑制。结论:南方红豆杉枝叶及其愈伤组织萃取紫杉醇后剩余部分对α-糖苷酶均有一定的抑制活性,愈伤组织较优于枝叶。
OBJECTIVE: To study the effects of the remaining fractions of Taxus chinensis and its callus on the activity of α-glucosidase after its extraction. Methods: The remaining fractions of paclitaxel were added into different trace reaction systems of α-amylase, α-maltase and α-glucosidase, respectively. The enzyme activity was determined by Bernfeld method, PNPG method and GOD method, respectively. Graph method to study its kinetic properties. Results: Foliar and its callus had significant inhibitory effect on α-maltase and α-glucosidase, which was consistent with the effect of Baitangping, which inhibited the existence of dose-effect relationship and inhibited α-maltase activity [IC50 = ( (IC50 = (7.99 ± 1.57) mg / mL and (19.87 ± 2.52) mg / mL, respectively) were significantly higher than that of the control group (218.49 ± 18.24 mg / mL and 91.04 ± 5.43 mg / mL] / mL] showed irreversible, competitive inhibition. CONCLUSION: The remaining fractions of Taxus chinensis and its calli after extracting paclitaxel have certain inhibitory activity on α-glucosidase, and the callus is superior to the branches and leaves.
