Rapid analysis of airborne pathogens play a critical role in early warning of spreading of infectious diseases,which is essential for public health and disease prevention.Herein we developed series of integrated microfluidic devices that can execute airborne pathogen capture,enrichment and continuous-flow high-throughput accurate bioanalysis.Pathogens,including Klebsiella pneumoniae,Citrobacter koseri,Staphyloccocus aureus,Escherichia coli,Pseudomonas aeruginosa,Enterococcus faecalis and Mycobacterium smegmatis,have been used to validate the capture and analysis efficiency of the system.The experimental results showed that capture efficiency and detection limit had been improved with these microfluidic devices compared with the traditional method.The capture efficiency of microfluidic chip reaches almost 100%,and the detection limits down to approximately 118 cells were achieved toward E.coli,without the DNA purification process.It can collect enough bacteria from low concentration bioaerosol for the downstream direct protein/nucleic acid analysis.The whole system is rigid and simple,perfect for field application,the hospital or other high-risk airborne pathogen environment,showing potential application in environmental monitoring and public health protection.
