Background Recent work on genome-wide association studies (GWASs) in Caucasian and Han populations had discovered several novel susceptibility genes for psoriasis. Objective To detect the association of risk loci for psoriasis identified in the previous GWAS in Caucasian population with Chinese Uygur population in Xinjiang. Methods The genotyping analysis of 8 psoriasis associatated SNPs was performed in 539 psoriasis patients and 749 controls of Chinese Uygur using the Sequenom MassArray system (Sequenom iPLEX assay). Results Among 8 SNPs,two were confirmed to be associated with Chinese Uygur psoriatic group (SNP rs495337 at ZNF313: P=4.0*10-3,OR=0.7917; SNP rs20541 at IL13: P=2.1*10-3,OR=0.82).Subgroup analysis showed that MAF of the two SNPs significantly associated with type I psoriasis patients (p<0.05).rs20541 might preferentially associated with psoriasis in early age of onset whereas the ratio of negative and positive family history of psoriasis was not statistically different.Interestingly,a significant 2-locus interaction was observed by MDR between SNP rs495337 and rs20541 with a cross-validation consistency of 4/5 and average balanced prediction (accuracy=55.5%,p<0.001).Conclusions ZNF313 and IL13 are not only associated with psoriasis in the Chinese Uygur population but also have a interacted effect on conferring risk to psoriasis of Chinese Uygur population.
