Objectives: The main objective of this study was to determine the association beween antibiotic resistance and mobile resistance determinants including integrons and plasmid-mediated quinolone resistance(PMQR)genes in Proteeae strains collected from inpatients at the First Affiliated Hospital of Wenzhou Medical University.Methods: Class 1/2/3 integrons and qnrD were amplified by polymerase chain reaction(PCR)in 176 non-duplicate clinical isolates of proteeae.Antimicrobial susceptibility was determined by the agar dilution method and other PMQR determinants were also determined by PCR.Integrons gene cassettes were identified by restriction fragment length polymorphism and DNA sequencing analysis.Results: Of the 176 Proteeae strains,68.2%(120/176)Proteeae isolates exhibited multidrug resistance(MDR)phenotypes.PCR screening showed that 81(46.0%)and 19(10.8%) Proteeae isolates were positive for intI1 and intI2,respectively.No isolates carried intI3.Susceptibility testing showed that the resistance rate of integron-positive strains to aminoglycosides,fluoroquinolones,furantoin,and trimethoprim/sulfamethoxazole were significantly higher than integron-negative strains.Moreover,integron presence was associated with increased resistance range in terms of both the number of antimicrobials and the number of classes of antimicrobials to which a strain was resistance.Sequence analysis revealed fifteen different cassette arrangements were identified within class 1 integrons: aadB-catB3,dfrA12-aadA5,dfrA12-orfF-aadA2,dfrA17-aadA5,cmIA1-aac(6)-Ib-aacA4,dfrA17,aadB-qacEdelta1,aadB-cat-blaOXA10-aadA1,dfrA1-orjC,aadA1,aadA2,blaPSEI,aadB-aadA2,dfrA15 and aadB-cat-blaOXA10-aadA1-dfrA1-aacA4.Four different arrangements in class 2 integron: dfrA1-sat2-aadA1,dfrA1-catB2-sat2-aadA1,sat2-aadA1 and dfrA12-sat2-aadA1.PMQR was determined by qnrD in forty strains,and 14 strains were found to co-produced other resistance genes: aac(6)-Ib-cr(8 isolates),qnrB(2 isolates),qepA(3 isolates)and oqxAB(5 isolates).Conclusion: Our results indicate that class land 2 integrons are common among MDR isolates and can be used as a marker of multiresistance.Meanwhile,qnrD are highly prevalent in Proteeae isolated from our hospital.
