A small conductance Ca2+-activated K+ (SK) channels, SK2 channel, is important in configuration of action potential in atrial myocytes.Here, we determined the potential role of RyRs on SK2 channel.Methods:RyR2 mRNA level in cultured neonatal myocytes transfected with lentiviral-mediated small hairpin interference RNA (shRNA) against RyR2 was analyzed by real-time PCR.Whole cell patch-clamp technique was used to record Ca2+-activated K+ current (IK, Ca) in isolated mouse atrial myocytes Results: Four siRNAs reduced level of RyR2 mRNA in neonatal mouse myocytes up to 85%.The adult mouse atrial cells transdued with lentivirusmediated with shRNA specific to RyR2 showed a significant decrease the IK.Ca.Our study identifies that RyR2 plays an important role in the SK2 channel in mouse cardiac myocytes.
