A Gram-reaction-negative,aerobic,non-motile,rod-shaped and yellow-pigmented bacterial strain LPB0003T,was isolated from a Thais luteostoma in Yellow Sea,Korea.The phylogenetic analysis based on 16S rRNA gene sequence placed the test strain within the genus Polaribacter,a member of the family Flavobacteriaceae.The highest sequence similarity was observed with Polaribacter reichenbachii(97.8%)and followed by P.dokdoensis(97.7%),P.atrinae(97.4%),and P.irgensii(97.1%).Other Polaribacter species showed less than 97%similarity.Though strain LPB0003T showed high sequence similarity with its close neighbors,it did not cluster with any of the relatives.Rather,it formed a distinct phyletic clade within the radiation of Polaribacter.Optimal growth occurred in the presence of 2%(w/v)NaCl,at 25-27℃ and at pH 6.5.Oxidase-and catalase-activities were positive.According to API ZYM gallery,esterase(C4),esterase lipase(C8),leucine arylamidase,cystine arylamidase(weakly),valine arylamidase,trypsin,alkaline phosphatase,acid phosphatase,naphthol-AS-BI-phosphohydrolase,a-glucosidase(weakly),and N-acetyl-β-glucosaminidase were positive.In API 20NE strips,nitrate reduction and esculin hydrolysis were positive.The phenotypic characteristics distinguished strain LPB0003T from the close neighbors.The genome sequencing is under processing to determine the genomic relatedness among the test strain and reference strains.The taxonomic position of strain LPB0003T is still under investigation with the strong possibility that the test strain is a novel species of the genus Polaribacter.
