Background:Non-response of HCV patients to combination therapy has previously been associated with a strong hepatic up-regulation of interferon stimulated genes (ISGs) including ISG 15.Therefore,the aim of this study was to further elucidate the functional role of this molecule.Methods:ISG15 expression was suppressed by siRNAs or enhanced by over-expression in genomic and subgenomic human or murine HCV replicons systems.In addition,ISG 15 expression was analyzed in liver samples of HCV patients prior to antiviral therapy and correlated with clinical and virological parameters.
