目的:研究四逆散对实验性溃疡性结肠炎(UC)大鼠细胞因子mRNA表达的影响。方法:将实验动物分为5组,免疫法造模,采用Q-PCR技术检测实验大鼠结肠组织IL-1β、TNF-α、IL-4和IL-10 mRNA的表达。结果:模型组IL-1β(7.84±1.18)、TNF-α(2.45±0.19)mRNA的表达均明显高于正常组,而IL-4(0.34±0.09)、IL-10(0.59±0.08)mRNA表达明显低于正常组,P<0.01;与模型组比较,四逆散组、柴芍枳组和柴芍组IL-1β、TNF-αmRNA表达均显著降低,IL-4、IL-10 mRNA表达则显著升高,P<0.01;与四逆散组比较,柴芍枳组TNF-α(1.37±0.12)mRNA表达明显高于四逆散组(1.08±0.03)(P<0.05),柴芍组(0.79±0.03)明显低于四逆散组(P<0.05),柴芍枳组(1.09±0.05)(P<0.05)和柴芍组(1.33±0.15)(P<0.01)IL-4 mRNA表达均显著升高。结论:四逆散干预实验性UC的作用机理可能为抑制促炎因子IL-1β的移行和TNF-α的分泌,促进抗炎因子IL-4和IL-10的分泌,调节促炎因子和抗炎因子之间的平衡,方中君臣配伍作用尤为突出,占有重要地位。
Objective: To investigate the effect of Sini Decoction on cytokine mRNA expression in experimental ulcerative colitis (UC) rats. Methods: The experimental animals were divided into 5 groups. The immune system was used to establish the model. The mRNA expression of IL-1β, TNF-α, IL-4 and IL-10 in the colon of experimental rats was detected by Q-PCR. Results: The mRNA expression of IL-1β (7.84 ± 1.18) and TNF-α (2.45 ± 0.19) in the model group were significantly higher than those in the normal group, while the expressions of IL-4 (0.34 ± 0.09) and IL-10 Compared with the model group, the expression of IL-1β and TNF-αmRNA in Sini Decoction group, Chaihao Zhizhu group and Chaishaohu group were significantly decreased, the expressions of IL-4 and IL-10 mRNA were significantly decreased (P <0.05). Compared with Sini Decoction group, the expression of TNF-α (1.37 ± 0.12) mRNA in Chaishao Zhideng group was significantly higher than that of Sini powder group (1.08 ± 0.03) (P <0.05) (0.79 ± 0.03) was significantly lower than those of Sini Decoction (P <0.05), Chaihao Huzhi group (1.09 ± 0.05) and Chaishao group (1.33 ± 0.15) (P <0.01) mRNA expression were significantly increased. CONCLUSIONS: The mechanism of Sini Decoction interfering with experimental UC may be to inhibit the migration of proinflammatory cytokines IL-1β and the secretion of TNF-α, promote the secretion of anti-inflammatory cytokines IL-4 and IL-10, regulate proinflammatory cytokines and anti- The balance between the inflammatory factors, Fang Zhongjun compatibility is particularly prominent, occupies an important position.
