基于人源性细胞的药物心脏毒性评价体外替代方法研究及应用

  目的 心脏毒性一直是新药研发失败的主要原因之一,其评价方法也是各国学者研究的热点.本文对基于人源性细胞的药物心脏毒性评价体外替代方法进行了研究.方法 采用转染hERG钾通道或Nav1.5钠通道、Cav1.2钙通道基因的HEK293细胞,利用膜片钳技术记录hERG钾电流或Nav1.5钠电流、Cav1.2钙电流,并采用阳性药物对上述电流进行验证;采用人胚胎干细胞分化的心肌细胞(human embryonic stem cell-derived cardiomyocyte,hESC-CM),接种到RTCA Cardio E-Plate 96孔板,利用实时细胞分析技术观察hESC-CM的搏动频率和幅度,并采用不同浓度的阳性药物进行验证;在此基础上,利用上述方法,评价化合物的心脏毒性.
