目的研究葡萄原花青素(GPC)对乙醇诱发小鼠肝细胞Caspase-3和Bax蛋白异常表达的影响。方法将小鼠随机分为正常对照组、乙醇对照组和各剂量GPC组,除正常对照组外,每组小鼠均每天经口灌胃乙醇4g/kg,低、中、高剂量GPC组同时分别给予GPC 50、100和200mg/kg,连续4周。取肝组织用MTT法检测各组小鼠的肝细胞增殖活性,用免疫组织化学法检测肝细胞Caspase-3和Bax蛋白的表达水平。结果高剂量GPC组Caspase-3和Bax蛋白阳性表达率分别为19.08%和14.06%,乙醇对照组则为51.78%和58.08%,差异有显著性(P<0.05)。高、中剂量GPC组细胞增殖活性高于乙醇对照组(P<0.05)。结论 GPC可抑制乙醇诱发的肝细胞Caspase-3和Bax蛋白异常表达,对乙醇性肝损伤具有防护作用。
Objective To investigate the effect of grape proanthocyanidin (GPC) on abnormal expression of Caspase-3 and Bax in ethanol-induced hepatocytes of mice. Methods The mice were randomly divided into normal control group, ethanol control group and each dose of GPC group, except for the normal control group, each group mice were orally administered with ethanol 4g / kg, low, middle and high dose GPC group The rats were given GPCs at doses of 100, 100 and 200 mg / kg for 4 weeks. The liver tissue was collected to detect the hepatocyte proliferation activity by MTT assay and the expression of Caspase-3 and Bax protein in hepatocytes by immunohistochemistry. Results The positive rates of Caspase-3 and Bax protein in high-dose GPC group were 19.08% and 14.06% respectively, while those in ethanol group were 51.78% and 58.08%, respectively. The difference was significant (P <0.05). The cell proliferation activity of high and medium dose GPC groups was higher than that of ethanol control group (P <0.05). Conclusion GPC can inhibit the abnormal expression of hepatic cells Caspase-3 and Bax induced by ethanol and has a protective effect on alcoholic liver injury.
