Objectives:Matrix metalloproteinase 1(MMP1) plays pivotal roles in both normal development and in numerous pathologies.However, it remains unclear whether MMP1 participates in the osteo/odontogenic differentiation of human stem cells from the apical papilla (SCAPs).Methods:In this study, SCAPs were isolated and purified from human third molars with developing roots.Meanwhile, retrovirus overexpression plasmid PMSCV/MMP1 was constructed.After transfection and selection, we established cell lines which stably expressed MMP1 (SCAPs-M).Result:We found that alkaline phosphatase (ALP) activity and mineralization capacity of SCAPs-M were decreased.Western bolt and real-time RT-PCR further confirmed that OSX, RUNX2 and DSP, important osteo/odontogenisis-related factors, were suppressed in SCAPs-M.Blocking NF-κB signaling pathway derepressed the osteo/odontogenic differentiation caused by overexpression of MMP1.Conclusions:Taken together, these data suggested that MMP1 can suppress the osteo/odontogenic differentiation in SCAPs possibly via NF-κB pathway.
