Solutes have been isolating by specific type of stationary phase featured by one type of force.Secondary force from the matrix, space arm, or ligand is considered to harm the process of separation for a long time.Efforts have been made to decrease the secondary force.However, mixed-mode chromatography has been developed to separate small solute, peptides, and proteins.It has been proved to have advantages of higher selectivity, high load capacity.Each protein molecule has hydrophobic, hydrophilic, and charge residues.Although the stationary phase of protein developed quickly from the agarose-based to polymer based, silica-based, even monolith, the separation mode of proteins is only considered as one of reversed-phase liquid chromatography, hydrophobic interaction chromatography (HIC), ion-exchange chromatography (IEC), affinity chromatography, and so on.Each of these modes is controlled by a certain type of force.
