AIM Andrographolide salt (YGT) was synthesized as a salt to increase its solubility in the injection.We have studied its pharmacological effects about antibacterial, antivirus, antlinflammatory and immunoregulation and evaluated its activity.METHODS Using cylinder-plate method on YGT antibacterial activity in vitro studied, and to study the protection effect of mice after infected Staphylococcus aureus and Bacillus dysenteriae ; in vitro and in vivo experimental study of the antiviral, YGT on viral suppression and the characteristics of the infection of influenza virus in mice.The anti-inflammatory activities and the mechanisms of YGT were evaluated by using xylene-induced ear edema experiments, leukocyte migration experiments, and egg white in rat paw edema model,carrageenin-induced peritonitis experiments.The immune model that injecting chicken red cells in enterocoeha of mice and study the effect of YGT on immune function, taking Yanghuning(YHN) as the positive control.RESULTS In vivo and in vitro bacteriostasis experiments showed that, YGT has a weaker antibacterial action, no bactericidal effect, but can reduce the infection of Staphylococcus aureus mortality in mice in vivo, has good resistance of Staphylococcus aureus in vitro antiviral effect; experimental results show, YGT only very weak in vitro antiviral action, performed for the treatment of, directly kill virus or preventive effect; in vivo, at the dose of 125 mg·kg-1 on influenza virus infection in mice has extended the survival time and improve the rate of death protection role, at dose of 125 mg·kg-1 injection on pulmonary inflammation in mice have reduced effect; anti-inflammatory experiments show that, YGT and YHN used at the dose of 125 and 31 mg·kg-1 prevented xylene-induced ear dose significantly potassium sodium dehydroandrographolide succinate edema in normal mice.CMC-Na reduction caused by airbag in leukocyte extravasation, at 88 and 22 rag· kg-1 doses can reduce egg albumin induced rat paw swelling, In addition, YGT and YHN inhibited leukocyte reduced IL-lon migration, level in inflammatory exudates in serum.Immune tests show that, YGT can increase in spleen, thymus weight, increased in mice peritoneal macrophage phagocytic rate, with enhanced immune function.CONCLUSION (①) The result of bacteriostasis and antiviral test show that YGT is weak antibacterial antiviral effect in vitro.In addition, in vivo research , YGT has obvious protective effect on infected mice.② YGT and YHN showed significant anti-inflammatory activities on inhibiting leukocyte migration and reducing cytokines and mediators.YGT also has a strong effect on improving immunity and then can improve the mice against Staphylococcus aureus and influenza virus ability.The effect of YGT and YHN are well-matched , but YGT shows better activity in anti-inflammatory compared to YHN.
