Recurrent gene fusions and translocations were previously thought to be a mechanism relegated to hematological malignancies and rare soft tissue tumors (sarcomas), but our study changed this paradigm and suggests that this mechanism is important in common solid tumors (the carcinomas).We have observed recurrent fusions of the androgen-regulated gene TMPRSS2 with the ETS family of oncogenic transcription factors in the majority of prostate cancers (Science 28 October 2005).In general, when a recurrent gene fusion is identified for a particular cancer type, it is thought to be the causative or initiating molecular lesion.Thus, we try to uncover these gene fusions in common solid tumors as this will have profound implications the diagnosis and rational therapy for cancer.Here we report a strategy to systematically discover novel gene fusions in cancer cells using high throughput transcriptome sequencing platforms that integrate long read, short read and pair-end read.
