Cervical cancer is common gynecological malignancy in women worldwide.Growing evidence has shown that long non-coding RNAs(lncRNAs)can serve as biomarker in patients with cervical cancer.However,prognostic biomarkers for cervical cancer are still lacking.The objective of this study is to identify the lncRNA signature that could evaluate the prognosis in cervical cancer based on the data from The Cancer Genome Atlas(TCGA).The associations between survival outcome and the expression of lncRNAs were evaluated by the univariate and multivariate Cox proportional hazards regression analyses.We constructed a risk score as a method to evaluate prognosis.With validation,the expression levels of 2 key lncRNA in 31 newly diagnosed CC patients by qRT-PCR.There were 289 RNA sequencing profiles and the clinical data were obtained.A total of 49 differentially expressed lncRNAs were identified,among which 2 lncRNAs related to overall survival(OS)in CC patients.These two lncRNAs(ILF3-AS1 and RASA4CP)were found as a single prognostic signature.Meanwhile,CC patients with low-risk scores were more positively correlated with OS(P<0.001).Further analysis suggested that the prognostic value of this two-lncRNAs signature was independent in clinical features.Gene enrichment analysis found that these two lncRNAs were correlated with several molecular pathways.Results of qRT-PCR showed that the expression level of 2 lncRNAs were downregulated,the fold-changes between qRT-PCR and TCGA were consistent.In addition,the area under the ROC curve of 2 lncRNAs in CC were 0.704 and 0.685,(p <0.05).The AUC of the combine two lncRNAs was 0.723(p <0.05).Our study indicates that the two-lncRNAs signature could use as an independent biomarker for prognosis of cervical cancer,based on bioinformatics analysis.
