Objective TO investigate the effects of Tao Xin Fang on the learning and memory ability,and the synapse structure and neuronal structure and the number of synapses in hippocampal CA1 zoon of APP/PS1 transgenic AD mice.Methods Fifty-four three-month-old APP/PS1 double transgenic AD mice were randomly divided into model group,Tiao Xin Fang group and donepezil HCL group,with the same months of age and sex,in18 CS7BL / 6 j wild mice as normal control group.Lavage after 12 weeks,Morris water maze test was used to test behavior and Transmission electron microscopy was used to observe the synapse and neuronal structure of hippocampal CA1 zoons in mice.Results Compared with the normal group,model group mice escape latency were significantly prolonged (P<0.01),the number of crossing the platform was significantly reduced (P<0.05),passing through the original platform quadrant was significantly reduced (P<0.01);The neurons nuclear in hippocampus CA1 zoons were pyknosis,nuclear membrane was partially shrunk and thickened.tThere was an empty halo around the nucleus.and some of near the cell membrane were stained deeply,There are heterochromatin and swollen mitochondria.endoplasmic reticulum were vesicle shape.The number of synaptic neuropil was significantly reduced (P<0.01),synaptic structures appear large vacuoles.Compared with the model group,The tiaoxinfang grop escape latency were shorter (P<0.01),the number of crossing the platform was significantly increased (P<0.01),passing through the original platform quadrant was significantly increased (P<0.01).The neurons nuclear in hippocampus CA1 zoons were round,clear membrane was clear and complete,nucleolus was obvious;chromatin was fine and smooth,mitochondrial cristae was clearly visible.Mitochondria was partly swollen.The endoplasmic reticulum and free ribosomes were normal.The synaptic structure was obvious,the number of synapses was significantly increased (P<0.01),and the organelles lesion was reduced.Conclusion TiaoxinFang can improve the ability of learning and memory,and improve the degeneration of neuron and synapse structure and reduce the number of synapses.It has a protective effect on cognition and synapse.and and has a good prospect in the prevention and treatment of Alzheimers disease.
