In mammals,the pool of resting primordial follicles serves as the source of developing follicles andestablishment of this oocyte pool is essential for fertility.Mammalian primordial germ cells form and migrate to thegonad during embryonic development.After arriving at the gonad,the germ cells begin to mitosis forming clusters ofcells called germ line cysts or oocyte nests.Three days after birth,the oocyte nests break apart into individual cellsand become into primordial follicles.During this time,only part of oocytes ultimately survive and most of otheroocytes die by programmed cell death or else.Studies about the mechanisms have been continued for decades.Inrecent years,molecular mechanisms have become more evident,expecially in epigenetic.Epigenetics refers to acollection of mechanisms and phenomena that define the phenotype of a cell without affecting the genotype.Theepigenetic modify of germ cells,which is included DNA methylation,histone modifications and other changingdynamically during their development controls the stability expression and activation of genes closely.In this Review,we discuss dynamic epigenetic changes of Lhx8 and its expression that occur during mammalian germ-celldevelopment.Our results demonstrated that Lhx8 transcript localizes to the germ cell nucleus.In females,Lhx8expression commences as early as E13.5,when oocytes enter meiosis I during embryonic developmen and is highlyexpressed in primordial follicle formation.In addition,The interrelationship between the expression of Lhx8 andepigenetic modify is studied.We found that DNA methylation and histone acetylation is negativecorrelation to its expression. In a conclusion,Lhx8 plays an importment role during the formation of primordialfollicles,and this process is regulated by epigenetic.
